Tetracycline Programs
Tetracyclines were discovered more than 50 years ago and were for nearly four decades one of the best and
most used broad spectrum class of antibiotics. Worldwide, tetracyclines enjoy substantial use in treating
some of the most difficult bacterial infections, and physicians continue to use the class because of its safety,
tolerability and continued efficacy in selected indications. However, like many of the most established
antibiotic classes, the broad use of the tetracyclines has declined in recent decades because of the emergence
of bacterial resistance to these drugs.
Through the development of specialized techniques to synthesize diverse new tetracyclines and keen understanding
of the mechanisms of resistance, Paratek is developing improved compounds that overcome tetracycline and multi-drug
resistance (efflux and ribosomal protection are the two clinically relevant forms of resistance that affect
tetracycline antibiotics).
